For the most you should always try and optimise the best you can without using any "tools". Take a good look at some of the links below as a reference .

Great On Page Articles of Note

Baring in mind the first link has been updated this year. The banklinko article is pretty comprehensive and well worth a read.

After that point its always a good idea to use tools to make sure you didnt miss anything, which is easily done if you move to the more technical side of on-page seo with redirects, robots etc.

Some of the tools that are best appreciated for website audits

There are obviously many more but those particular ones I use practically daily.

Use different online tool , like to use moz one good one to refer to. For other features like speed Gtmatix is good one. Few other sites also give good feedback like one well know domain is neilpatel.

Either you can observe if things are changing depending upon the logged in user (e.g. every user sees different feeds on her Facebook wall) OR You can use a tool like BuiltWith to find out what kind of server side technologies are being used and make an intelligent guess.
